Etymology: portillorum: Named for D. and H. Portillo, who guided W.A.M. to the type locality and provided valuable natural history information for the area. The common name, Ulúan killifish, refers to the drainage in which it was found (Ref. 84033).

Dorsal soft rays (total): 10 - 15; Anal soft rays: 9 - 14. Profundulus portillorum belongs to the P. labialis species group and can be distinguished from its congeners by having a smaller eye and fewer anal and pectoral-fin rays. Eye diameter typically less than one fourth of the head length (greater than 25% in all other Profundulus), 12 anal fin rays modally (≥ 13 in all other Profundulus), 14 pectoral-fin rays modally (≥ 15 in all other Profundulus). It differs from P. guatemalensis, P. oaxacae and P. punctatus by the absence of a humeral spot and by having less than half of the basal portion of the caudal fin densely covered by scales. It is distinguished from P. hildebrandi and P. labialis by having <35 lateral-line scales and from P. candalarius by having a shorter body depth (Ref. 84033).

The specimens were collected from two sites at 1340 m elevation in two small tributaries of the Río Calam in the Ulúa drainage. The creeks were 3-5 m wide, 20-40 cm deep and canopy cover was c. 60%; substratum dominated by large cobble, and creek margins featured emergent macrophytes where the species was collected. Associated fishes Alfaro huberi, Oreochromis niloticus and Rhamdia laticauda (Ref. 84033).
